01 Running thesis

A Phase 3 bet on immune reset

Nektar is now mostly a bet on REZPEG. The drug is designed to boost regulatory T cells, which are immune cells that help calm the body when the immune system is overactive. In atopic dermatitis, a serious form of eczema, Phase 2b data showed meaningful skin improvement and durability on less frequent dosing.

The bull case is clear. Nektar has started ZENITH-AD, a global Phase 3 program in moderate-to-severe atopic dermatitis. The company says first topline Phase 3 data are expected in mid-2028, with a planned FDA biologics license application in 2029 if the data work. REZPEG also showed statistically significant improvement in asthma control scores among atopic dermatitis patients with asthma, which could matter because DUPIXENT is one of the few drugs known for helping across related allergic diseases.

Alopecia areata adds upside with a newly clarified path. After an end-of-Phase 2 meeting, Nektar reached FDA alignment on a single 850-patient registrational Phase 3 study named ZENITH-AA. The company plans to start this trial in early 2027.

The bear case is timing and competition. Nektar must run large trials, compete with well-known biologics, and convince doctors to switch or add a new immune approach. The balance sheet gives it time, but the valuation score is weak because investors are being asked to price in success years before any possible commercial launch.

02 Business model

Cash funds trials, not sales

Nektar does not yet have a normal product-sales business. It is a clinical-stage research company, so its value comes from drug candidates moving through trials and, later, possible approvals or partnerships. Today, the key spending need is Phase 3 development for REZPEG.

The company regained full rights to REZPEG after Eli Lilly ended their collaboration. That gives Nektar more control and more upside if the drug works, but it also means Nektar must carry more of the cost and execution risk itself.

The funding picture is currently stable. Nektar ended the second quarter of 2026 with $1.02 billion in cash and investments. Management expects to end 2026 with $815 million to $840 million in cash. This extends the financial runway into the third quarter of 2028.

That cash cushion lowers near-term financing risk, but it came through heavy equity issuance over the prior year. Existing shareholders were diluted. The main business question is whether the upcoming data can create enough value to justify that dilution.

ZENITH-AD fails or looks too ordinary

The main value driver is REZPEG in atopic dermatitis. If Phase 3 does not confirm the Phase 2b benefit, or if safety looks worse in larger trials, the stock thesis would change fast. Even a positive trial could disappoint if the effect does not look strong enough versus established drugs.

Doctors stay with entrenched biologics

DUPIXENT is already a major atopic dermatitis drug and has a strong position with doctors. REZPEG may need to prove durability, convenient dosing, and benefit in related conditions like asthma to win share. A new mechanism alone is not enough if doctors see no clear reason to switch.
